When the isotonic low ionic strength medium was supplemented with 25mM KCl, a several-fold increase in the succinate-ferricyanide reductase activity of mitochondria and in the rate of external NADH oxidation in the presence of ferrocyanide was observed. Mitochondrial respiration with 5mM ferrocyanide was almost completely inhibited after consumption of 3.8-18.5% of the dissolved oxygen, depending on the medium and the presence of 2,4-dinitrophenol. Departamento de Ciencias; Ciencias Biológicas y Bioprocesos (CIBIOP)Bacillus subtilis EA-CB0015 was isolated from the phyllosphere of a banana plant and tested for its potential to produce bioactive compounds against Mycosphaerella fijiensis. Using a dual plate culture technique the cell-free supernatant of B. subtilis EA-CB0015 produced inhibition values of 89 +/- 1%. The active compounds were purified by solid-phase extraction and HPLC, and their primary structures determined using mass spectrometry and amino acid analysis. A new fengycin isoform, fengycin C, with the amino acid sequence Glu-Orn-Tyr-Thr-Glu-Val-Pro-Gln-Thr-Ile was isolated. The peptidic moiety differs from fengycin B at position 9 and from fengycin A at positions 6 and 9. The beta-hydroxy fatty acyl chain is connected to the N-terminal of the decapeptide and can be saturated or unsaturated, ranging from 14 to 18 carbons. Departamento de Ciencias; Ciencias Biológicas y Bioprocesos (CIBIOP)Culture conditions in shake flasks affect filamentous Streptomyces lividans morphology, as well the productivity and O-mannosylation of recombinant Ala-Pro-rich O-glycoprotein (known as the 45/47 kDa or APA antigen) from Mycobacterium tuberculosis. In order to scale up from previous reported shake flasks to bioreactor, data from the literature on the effect of agitation on morphology of Streptomyces strains were used to obtain gassed volumetric power input values that can be used to obtain a morphology of S. lividans in bioreactor similar to the morphology previously reported in coiled/baffled shake flasks by our group. Morphology of S. lividans was successfully scaled-up, obtaining similar mycelial sizes in both scales with diameters of 0.21 ± 0.09 mm in baffled and coiled shake flasks, and 0.15 ± 0.01 mm in the bioreactor. Moreover, the specific growth rate was successfully scaled up (0.09 ± 0.02 and 0.12 ± 0.01 h(-1), for bioreactors and flasks, respectively), and the recombinant protein productivity measured by densitometry, as well. More interestingly, the quality of the recombinant glycoprotein measured as the amount of mannoses attached to the C-terminal of APA was also scaled- up; with up to five mannose residues in cultures carried out in shake flasks; and six in the bioreactor. Departamento de Ciencias; Mauricio A TrujilloRoldán (maurotru@gmail.com); Ciencias Biológicas y Bioprocesos (CIBIOP)The impact of flask geometry on Streptomyces lividans growth and morphology, production and Omannosylation of a recombinant O-glycoprotein (APA from Mycobacterium tuberculosis) was described and associated to the evolution of the volumetric power input (P/V) in three shake flask geometries -- During the exponential growth, the highest P/V was found in baffled flasks (BF) with 0.51 kW/m3, followed by coiled flasks (CF) with 0.44 kW/m3 and normal Erlenmeyer flasks (NF) with 0.20 kW/m3 (flasks volume of 250 mL, filling with 50 mL and agitated at 150 rpm) -- During the stationary phase, P/V decreased 20% in BF and CF, but increased two times in NF, surely due to changes in mycelial morphology and its effects on rheology -- Also, NF cultures were carried out at a filling volume and agitation of 15 mL, 150 rpm (15 mL-NF), and 25 mL, 168 rpm (25 mL-NF), in order to raise P/V closely to the values obtained in CF -- However, different growth, morphology and recombinant protein productivity were obtained -- These data indicate that P/V is not a definitive parameter that can determine bacteria growth and morphology, not even glycoprotein production -- But it can be proposed that the oxygen transfer in the center of the pellets and hydromechanical stress might be the more relevant parameters than P/VÍtem Multiple response optimization of Bacillus subtilis EA-CB0015 culture and identification of antifungal metabolites(Biocatalysis and Agricultural Biotechnology, 2014) Ordúz, Sergio; Mosquera, Sandra; González Jaramillo, Lina María; Villegas Escobar, Valeska; Escuela de Biociencias; Centro de Investigaciones del Banano; Universidad EAFIT. Departamento de Ciencias; Valeska Villegas-Escobar (vvilleg2@eafit.edu.co); Ciencias Biológicas y Bioprocesos (CIBIOP)The low yields of biomass and antimicrobial metabolites obtained in fermentation processes are limiting factors for implementing biological control agents in the field -- In this context, optimization of the culture medium for the biological control agent Bacillus subtilis EA-CB0015 was conducted in submerged culture to maximize the biomass production and antifungal activity -- Additionally, the active metabolites against the phytopathogen Mycosphaerella fijiensis produced under optimized conditions were identified -- Carbon and nitrogen constituents of the culture medium were optimized using a 22 factorial design with central point followed by a multiple response optimization coupled to the desirability function approach -- The optimized medium (33.4 g/L of glucose and 32.5 g/L of yeast extract), showed significant increases in both, cell density by 3.6 folds (13.271.0 g/L) and antifungal activity by 1.2 folds (77.770.3% inhibition) -- To gain insight into the type of active compounds, they were purified and identified by mass spectrometry (MS) -- The MS analysis revealed the presence of three families of lipopeptides: surfactin, fengycin and iturin -- The high antifungal activity was associated with the novel fengycin C and with iturin -- A which were partially growth-associated with a maximum concentration of 781.4 and 355.4 mg/L at 36 h of growth, respectively -- These results indicate the potential of B. subtilis EA-CB0015 to produce high concentrations of biomass and lipopeptides that can be exploited for biotechnological applicationÍtem Fengycin C produced by Bacillus subtilis EA-CB0015(American Chemical Society, 2014) Ceballos, Isabel; J. ( gsantafe@correo.unicordoba.edu.co); Ciencias Biológicas y Bioprocesos (CIBIOP)This study reports the synthesis of five (E)-Styrylquinolines (Q.1, Q.2, Q.3, Q.5, Q.6) and three O-alkylated derivatives (Q.1a, Q.6a, Q 6 b) and the evaluation of its antimalarial activity and cytotoxic in vitro -- For Styrylquinolines Perkin condensation reaction type from 8-hydroxyquinaldine with aromatic aldehydes was used -- Antimalarial activity was performed using the method Radioisotope with FCB-2 strains of Plasmodium falciparum (chloroquine-resistant) and for the cytotoxic activity the MTT method with HepG2 cells was employed -- Most compounds showed to be active antimalarial -- Those with higher activity were Q.3 (IC506.4μM) and Q.5 (IC5010.8 µM), although all of them were cytotoxic at concentrations above 1 µg / mL -- The structures of these compounds were confirmed using infrared spectroscopy techniques and MRI in one and two dimensions
