Given its genomic diversity, the strain Bacillus subtilis EA-CB0575, isolated from the rhizosphere of a banana plant, was sequenced and assembled to determine the genomic potential associated with its plant growth promotion potential. The genome was sequenced by Illumina technology and assembled using Velvet 1.2.10, resulting in a whole genome of 4.09 Mb with 4332 genes. Genes involved in the production of indoles, siderophores, lipopeptides, volatile compounds, phytase, bacilibactin, and nitrogenase were predicted by gene annotation or by metabolic pathway prediction by RAST. These potential traits were determined using in vitro biochemical tests, finding that B. subtilis EA-CB0575 produces two families of lipopeptides (surfactin and fengycin), solubilizes phosphate, fixes nitrogen, and produces indole and siderophores compounds. Finally, strain EA-CB0575 increased 34.60% the total dry weight (TDW) of tomato plants with respect to non-inoculated plants at greenhouse level. Departamento de Ingeniería de Producción; Materiales de IngenieríaAmong many dermal armors, fish scales have become a source of inspiration in the pursuit of “next-generation” structural materials. Although fish scales function in a hydrated environment, the role of water and intermolecular hydrogen bonding to their unique structural behavior has not been elucidated. Water molecules reside within and adjacent to the interpeptide locations of the collagen fibrils of the elasmodine and provide lubrication to the protein molecules during deformation. We evaluated the contributions of this lubrication and the intermolecular bonding to the mechanical behavior of elasmodine scales from the Black Carp (Mylopharyngodon piceus). Scales were exposed to polar solvents, followed by axial loading to failure and the deformation mechanisms were characterized via optical mechanics. Displacement of intermolecular water molecules by liquid polar solvents caused significant (p = 0.05) increases in stiffness, strength and toughness of the scales. Removal of this lubrication decreased the capacity for non-linear deformation and toughness, which results from the increased resistance to fibril rotations and sliding caused by molecular friction. The intermolecular lubrication is a key component of the “protecto-flexibility” of scales and these natural armors as a system; it can serve as an important component of biomimetic-driven designs for flexible armor systems. Statement of Significance: The natural armor of fish has become a topic of substantial scientific interest. Hydration is important to these materials as water molecules reside within the interpeptide locations of the collagen fibrils of the elasmodine and provide lubrication to the protein molecules during deformation. We explored the opportunity for tuning the mechanical behavior of scales as a model for next-generation engineering materials by adjusting the extent of hydrogen bonding with polar solvents and the corresponding interpeptide molecular lubrication. Removal of this lubrication decreased the capacity for non-linear deformation and toughness due to an increase in resistance to fibril rotations and sliding as imparted by molecular friction. Departamento de Ingeniería de Procesos; Procesos Ambientales (GIPAB)The Response Surface Methodology (RSM) was applied as a tool for the optimization of the operational conditions of the photo-degradation of highly concentrated PY12 wastewater, resulting from a textile industry located in the suburbs of Medellin (Colombia). The Box-Behnken experimental Design (BBD) was chosen for the purpose of response optimization. The photo-Fenton process was carried out in a laboratory-scale batch photo-reactor. A multifactorial experimental design was proposed, including the following variables: the initial dyestuff concentration, the H(2)O(2) and the Fe(+2) concentrations, as well as the UV wavelength radiation. The photo-Fenton process performed at the optimized conditions resulted in ca. 100% of dyestuff decolorization, 92% of COD and 82% of TOC degradation. A kinetic study was accomplished, including the identification of some intermediate compounds generated during the oxidation process. The optimal values of the operation parameters, maximizing the coliform elimination percentage, were determined using statistical experimental design. The current density ( j: 2-20 mA/cm2), the conductivity (s: 500-900 µS/cm) and the anode materials (An) were considered as variables to perform the Box-Behnken experimental design together with the response surface methodology analysis for optimization. The statistical analysis indicated that, in the evaluated range, the disinfection efficiency increased with an increase in j and decreased with an increase in s. The following optimal conditions for the elimination of total and fecal coliforms were found: j: 10 mA/cm2, s: 500 µS/cm and BDD/Ti used as anode material. The BDD/Ti electrode let to achieve complete coliform elimination after ca. 20 min of reaction while the GP one needed ca. 27 min. In water treated with both BDD/Ti and GP anode, after 7 days, any coliforms growth was observed. Departamento de Ciencias; Ciencias Biológicas y Bioprocesos (CIBIOP)Bacillus subtilis EA-CB0015 was isolated from the phyllosphere of a banana plant and tested for its potential to produce bioactive compounds against Mycosphaerella fijiensis. Using a dual plate culture technique the cell-free supernatant of B. subtilis EA-CB0015 produced inhibition values of 89 +/- 1%. The active compounds were purified by solid-phase extraction and HPLC, and their primary structures determined using mass spectrometry and amino acid analysis. A new fengycin isoform, fengycin C, with the amino acid sequence Glu-Orn-Tyr-Thr-Glu-Val-Pro-Gln-Thr-Ile was isolated. The peptidic moiety differs from fengycin B at position 9 and from fengycin A at positions 6 and 9. The beta-hydroxy fatty acyl chain is connected to the N-terminal of the decapeptide and can be saturated or unsaturated, ranging from 14 to 18 carbons. Departamento de Ciencias; Ciencias Biológicas y Bioprocesos (CIBIOP)In this work, various ferro-ferricyanide oxidoreductase activities of rat liver mitochondria were studied to find conditions under which the outer membrane might restrict the flux of these highly charged non-biological anions. When the isotonic low ionic strength medium was supplemented with 25mM KCl, a several-fold increase in the succinate-ferricyanide reductase activity of mitochondria and in the rate of external NADH oxidation in the presence of ferrocyanide was observed. Mitochondrial respiration with 5mM ferrocyanide was almost completely inhibited after consumption of 3.8-18.5% of the dissolved oxygen, depending on the medium and the presence of 2,4-dinitrophenol. These and other experimental data together with mathematical modeling of the redox-state equilibrium suggest that the measured activities might be restricted by two factors: first, the permeability of the outer mitochondrial membrane and second, a strong influence of the ionic strength of incubation media on the intermembrane space redox reactions.Ítem The removal of the trivalent chromium from the leather tannery wastewater: the optimisation of the electro-coagulation process parameters.(IWA PUBLISHING, 2011-02-01) GilPavas, E.; Dobrosz-Gomez, I.; Gomez-Garcia, M. Departamento de Ingeniería de Procesos; Procesos Ambientales (GIPAB)The capacity of the electro-coagulation (EC) process for the treatment of the wastewater containing Cr3+, resulting from a leather tannery industry placed in Medellin (Colombia), was evaluated. In order to assess the effect of some parameters, such as: the electrode type (Al and/or Fe), the distance between electrodes, the current density, the stirring velocity, and the initial Cr3+ concentration on its efficiency of removal (%RCr+3), a multifactorial experimental design was used. The %RCr3+ was defined as the response variable for the statistical analysis. In order to optimise the operational values for the chosen parameters, the response surface method (RSM) was applied. Additionally, the Biological Oxygen Demand (BOD5), the Chemical Oxygen Demand (COD), and the Total Organic Carbon (TOC) were monitored during the EC process. The electrodes made of aluminium appeared to be the most effective in the chromium removal from the wastewater under study. At pH equal to 4.52 and at 28 degrees C, the optimal conditions of Cr3+ removal using the EC process were found, as follows: the initial Cr3+ concentration=3,596 mg/L, the electrode gap=0.5 cm, the stirring velocity=382.3 rpm, and the current density=57.87 mA/cm2. At those conditions, it was possible to reach 99.76% of Cr3+ removal, and 64% and 61% of mineralisation (TOC) and COD removal, respectively. Departamento de Ingeniería de Producción; Ingeniería, Energía, Exergía y Sostenibilidad (IEXS)The resolution of quaternary mixtures of chlorophylls a and b and pheophytins a and b has been accomplished by partial least-squares (PLS) multivariate calibration, applied to the fluorescence signals of these pigments. The total luminescence information of the compounds has been used to optimize the spectral data set to perform the calibration. After preliminary studies, a method is described in acetone media, to avoid emulsions with the olive oil samples. Different scanning paths have been selected for each method. For the simultaneous determination of the pigments in olive oil samples, a comparative study of the results found by using excitation, emission, and synchronous spectral data, as analytical signal, was performed. The excitation spectra were selected as the better analytical signals for the determination of the pigments in olive oil samples. The optimum wavelength range to record the excitation spectra (?em = 662 nm) was selected to minimize the contribution of pheophytin a and to maximize the contribution of the other pigments, which are the minor constituents in olive oil. Determination of these pigments in olive oil samples was effected from the excitation spectra of dissolutions o suitable aliquots in acetone. Recovery values from olive oil, spiked with chlorophylls a and b and pheophytins a and b, were in the ranges of 70-112, 71-111, 76-105, and 82-109%, respectively.
