Ítem Elaboración de un catálogo de bacterias cultivables como línea base de aislamientos con potencial hidrocarbonoclasta, de dos zonas con alta influencia petrolífera en Colombia(Universidad EAFIT, 2022) Enríquez Delgado, Sandra Milena; Pinel Peláez, NicolásFrom suspensions of environmental substrates (water, soil and sediment) from two zones with high oil influence in the country (Magdalena and Casanare) were isolated through conventional cultivation, 200 strains that formed the baseline of a catalog of bacterial biological interpretation. In this study, emphasis was placed on the cultivation process, isolation and integral identification (morphological, biochemical and genetic) of the isolates, with a view to understanding the repertoire and its functional dynamics. Molecular identification is based on genomic analysis from DNA sequencing and direct amplification of the 16s gene rRNA from bacterial biomass. Additionally, through the use of a minimal medium of salts supplemented with hydrocarbons at 1% v/v as the only carbon source, 27 were identified strains with hydrocarbonoclastic potential. The strains identified as tolerant to crude and bacterial consortia inoculated from soil samples directly, were tested of biodegradation, evaluating the percentage transformation of total hydrocarbons of oil (HTP) in 30 days of incubation. The biodegradation rates of HTP were determined under gas chromatography analysis coupled to a flame ionization detector (GCFID) and were related to the growth kinetics (CFU/mL) of the isolates through of the incubation time (30 days). Under observation time and post-identification molecular biodegradation of the isolates members of genera such as: Bacillus, Pseudomonas, Enterobacter and Burkholderia-Caballeronia-Paraburkholderia, in a range between 53.66 and 82% and in one of the consortiums 91.28%. The strains evaluated demonstrated an important enzymatic potential to degrade hydrocarbons and provide the opportunity to continue with the study of the bioremediation potential in areas affected by hydrocarbon contamination.
